## Overview and value chain

Markers: [EC: CDC/NIH BMBL 6th ed & WHO Lab Biosafety Manual 4th ed | OECD: Biotech & health | Regulator: FDA (USA), EMA (EU), NMPA (China)]

Biosafety and lab biosecurity is the physical-containment layer of biotechnology:
the cabinets, filtration, sterilization and room design that keep infectious
aerosols inside the laboratory and the product uncontaminated. The workhorse is
the Class II Type A2 biological safety cabinet (BSC), which draws room air through
a HEPA filter capturing 99.999 percent of particles at 0.3 micrometres, recirculates
about 70 percent and exhausts 30 percent through a second HEPA. Performance is set
by NSF/ANSI 49 in the United States and EN 12469 in Europe, while the four biosafety
levels (BSL-1 to BSL-4) of the CDC/NIH Biosafety in Microbiological and Biomedical
Laboratories (BMBL) manual and the WHO Laboratory Biosafety Manual dictate which
agent may be handled where. Sterilization closes the loop: autoclaves run a standard
121 °C, 15 psi gravity cycle for 15 to 30 minutes, with a 134 °C prion cycle for the
highest-risk material. The market is mature and concentrated in a handful of cabinet
and sterilizer makers — Thermo Fisher, Baker, Labconco, Getinge — whose installed
base underpins every pharmaceutical QC lab, diagnostics suite and high-containment
research facility.

The key directions of biosafety and lab biosecurity are:
1. **Class II biological safety cabinets (Class II BSC):** the dominant cabinet type
protecting personnel, product and environment via HEPA-filtered vertical laminar
airflow, specified under NSF/ANSI 49 and EN 12469 (Thermo 1300 Series, Baker
   SterilGARD, Labconco Logic).
2. **High-containment BSL-3/BSL-4 suites (BSL-3/4 Containment):** directional,
cascading negative-pressure room design with HEPA-filtered exhaust for respiratory
pathogens (BSL-3) and positive-pressure suits or gas-tight Class III cabinets for
   the highest-threat agents (BSL-4).
3. **Steam and sterilization technology (Steam Sterilization):** autoclaves and
   porous-load sterilizers running 121 °C / 134 °C cycles, plus vaporised
hydrogen-peroxide decontamination for cabinets and isolators (Getinge, Astell).
4. **Certification, maintenance and biosecurity (Certification & Biosecurity):**
annual NSF field certification of cabinet airflow and integrity, plus inventory,
access-control and select-agent compliance that turn containment into biosecurity.

## Tech stack and innovations

The biosafety stack rests on high-efficiency filtration, validated sterilization and
strict airflow geometry, each codified by decades of standards work.

1. **HEPA-filtered laminar airflow (HEPA Laminar Airflow):**
   - the Class II A2 cabinet draws room air through a HEPA (H14, 99.999 percent at 0.3 micrometres) and pushes a vertical laminar downflow over the work, recirculating about 70 percent.
   - a front grille captures the contaminated curtain and a second HEPA filters the 30 percent exhaust.
2. **Validated steam sterilization (Validated Steam Sterilization):**
   - porous-load autoclaves run a 121 °C, 15 psi cycle for 15 to 30 minutes for routine biohazard waste, with a 134 °C prion cycle for the highest-risk material.
   - cycles are validated by biological indicators (*Geobacillus stearothermophilus* spores) for GMP compliance.
3. **Directional negative-pressure containment (Negative-Pressure Containment):**
   - BSL-3 and BSL-4 suites use cascading pressure differentials of 10 to 15 pascals and HEPA-filtered exhaust so that air always flows from clean to contaminated zones.
   - BSL-4 adds positive-pressure suits or gas-tight Class III cabinets for agents such as Ebola and Nipah.

#### Stage 1: Airflow and cabinet design
The cabinet geometry is modelled so that a vertical laminar downflow of about 0.36 to 0.54 metres per second meets the front inflow, forming a protective air curtain at the work opening.

#### Stage 2: HEPA and downflow build
HEPA filters (H14, 99.999 percent efficient at 0.3 micrometres) are seated into the cabinet plenum, the blower and motor are installed, and the work surface, sash and lighting are fitted.

#### Stage 3: Factory certification
Each unit is tested at the factory to NSF/ANSI 49 (or EN 12469): downflow velocity, HEPA integrity by photometer scan, inflow velocity, site-accessibility and the KI-Discus protective test.

#### Stage 4: Field installation
The certified cabinet is placed away from doorways and air diffusers, connected via a thimble or hard-duct to the exhaust, and commissioned into the laboratory's HVAC and BSL pressure cascade.

#### Stage 5: Operation and decontamination
Daily work follows good microbiological technique; before filter changes or relocation the cabinet is decontaminated with vaporised hydrogen peroxide achieving a 6-log sporicidal kill.

#### Stage 6: Annual recertification
A field certifier re-tests downflow, inflow, HEPA integrity and airflow smoke patterns every twelve months, issuing the NSF recertification record that keeps the cabinet in regulatory compliance.
