Large-scale protein purification systems

Chromatography-based isolation of target proteins from complex mixtures, moving from batch Protein A capture to integrated and continuous trains — at manufacturing scale since 2022, with modular skids reaching about 8 tonnes a year.

1 · What it gives

Protein purification isolates target proteins from complex biological systems using their physical, chemical and structural properties, aiming at the required purity, concentration, activity and yield. The general process has three steps: pretreatment, rough fractionation and fine fractionation. Integrated and continuous purification systems are replacing traditional batch processing built on Protein A affinity chromatography; continuous downstream processes moved from proof of concept to manufacturing scale as of 2022. Integrated continuous plants using modular skids can reach annual outputs of up to 8 tonnes.

3 · How to implement 4 steps

  1. 1
    Pretreatment
    Prepare the feed (pretreatment).
  2. 2
    Rough fractionation
Capture and separate the bulk of the target (rough fractionation).
  3. 3
    Fine fractionation
Polish to the required purity, concentration, activity and yield (fine fractionation).
  4. 4
    Automate and comply
Run the train on an integrated automated system compliant with 21 CFR Part 11.
