Stage 1: Formulation and filling

The sterile bulk drug product is sterile-filtered and dosed into washed, depyrogenated vials that are only partially stoppered, leaving the vent open so that water vapour can escape during sublimation.

Stage 2: Freezing and nucleation

Vials are loaded, manually or by an ALUS system, onto the freeze-dryer shelves and frozen; with controlled nucleation (LyoCoN or Lyonuc) ice forms simultaneously across the batch near -4 °C instead of stochastically over 30-45 minutes, producing a uniform pore structure.

Stage 3: Primary drying (sublimation)

The chamber is pulled to deep vacuum, often below 0.15 mbar, and shelf temperature is raised; ice sublimes, transporting water vapour at sublimation rates exceeding 3 kg/m²h to the condenser (rated from 8 kg up to 800 kg) and removing the bulk of the water.

Stage 4: Secondary drying (desorption)

Shelf temperature is ramped under vacuum to desorb bound water, bringing residual moisture typically below 1% and maximising the shelf stability of the cake.

Stage 5: Stoppering and unloading
