## Overview and value chain

Markers: [EC: ASME BPE / USP <1043> / BPOG single-use standards | OECD: Industrial biotechnology | Regulator: FDA (USA), EMA (EU), NMPA (China)]

Wave bioreactors are single-use rocking platforms in which a pre-sterilised
flexible culture bag is rocked on an electrically driven base so that the
liquid travels as a periodic wave, mixing and aerating the culture without a
mechanical impeller or sparger. The format was introduced by Wave Biotech
(now Cytiva's ReadyToProcess WAVE and Xuri lines) and is the workhorse of
the N-1 seed train and early cell-expansion steps that feed every modern
mAb, vaccine and cell-therapy process: the ReadyToProcess WAVE 25 runs up to
a 25 L working volume in a gamma-irradiated Cellbag, Sartorius's Biostat RM
spans a 2–200 L portfolio (1–100 L working volume) on Flexsafe RM bags,
Thermo Fisher's HyPerforma Rocker runs Rocker BPCs to a 25 L working volume
under the G3 Lab Controller, and Applitech's 艾贝泰 AbioWave covers the Chinese
rocking-bioreactor market. Because there is no shaft seal or rotating impeller,
shear stays low enough for shear-sensitive stem, CAR-T and ATMP lines that
stirred tanks damage.

Key directions of wave bioreactors for seed-train expansion:
1. **Rocking wave/rocker single-use bioreactors (Rocking-Wave SUB):** the core
format — Cytiva ReadyToProcess WAVE / Xuri, Sartorius Biostat RM, Thermo Fisher
HyPerforma Rocker, Applitech AbioWave — rocking a Cellbag/Flexsafe RM bag at
   6–40 rpm for wave mixing.
2. **Low-shear cell-therapy and ATMP expansion (Low-Shear Cell-Therapy Expansion):**
rocker platforms such as Xuri optimised for stem-cell, CAR-T and
regenerative-medicine seed trains where impeller shear is prohibitive.
3. **Single-use Cellbag / Flexsafe RM formats (Single-Use Cellbag/Flexsafe RM):**
   gamma-irradiated 2–200 L EVA/UPE bags with integrated gas,
sampling and harvest ports that define the format's closed, sterile workflow.
4. **Wave-based perfusion and N-1 intensification (Wave Perfusion/Intensification):**
running the rocking bag in perfusion mode to densify the N-1 seed and cut the
   production bioreactor's turnaround time.

## Tech stack and innovations

The wave-bioreactor stack is built on rocking-wave hydrodynamics, single-use bag engineering and closed low-shear cell expansion.

1. **Rocking-wave mixing (Rocking-Wave Mixing):**
   - the platform rocks the bag at a controlled angle (6–10°) and rate (6–40 rpm), generating a travelling wave that mixes and aerates without an impeller or sparger.
   - case: Cytiva ReadyToProcess WAVE 25 and Sartorius Biostat RM.
2. **Single-use Cellbag / Flexsafe RM engineering (Single-Use Bag Engineering):**
   - gamma-irradiated EVA/UPE 3D bags with integrated gas, sampling and harvest ports, qualified to 25–40 kGy and SAL 10^-6 under USP <1043> and BPOG extractables/leachables.
   - case: Cytiva Cellbag and Sartorius Flexsafe RM.
3. **Low-shear cell-therapy expansion (Low-Shear Cell-Therapy Expansion):**
   - rocker platforms tuned for stem-cell, CAR-T and ATMP lines where impeller shear damages cells; integrated single-use pH/DO sensors and closed transfer to the production SUB.
   - case: Cytiva Xuri Cell Expansion System and Thermo Fisher HyPerforma Rocker.

#### Stage 1: Bag film & Cellbag fabrication
EVA/UPE multilayer film is extruded, welded into a 3D Cellbag with gas, sampling and harvest ports, and integrated with single-use pH/DO sensors and gas filters.

#### Stage 2: Gamma irradiation
The fabricated bag is gamma-irradiated to 25–40 kGy to a Sterility Assurance Level of 10^-6 and qualified for extractables and leachables under USP <1043> and the BPOG model.

#### Stage 3: Platform loading
The sterile Cellbag is loaded onto the wave/rocker platform (Cytiva WAVE/Xuri, Sartorius Biostat RM, Thermo Fisher HyPerforma Rocker, Applitech AbioWave), connected to the gas mixer and single-use sensors, and inoculated.

#### Stage 4: Inoculation & wave culture
Cells are seeded at 0.3–0.5 million cells/mL and the bag is rocked at 6–40 rpm and a 6–10° angle; the travelling wave mixes and aerates the culture without an impeller, sustaining CHO, HEK293, stem-cell and CAR-T expansion.

#### Stage 5: Scale-up & perfusion
The culture is expanded by fed-batch or perfusion to densify the N-1 seed — wave perfusion lifts cell density high enough to cut the production bioreactor's turnaround time.

#### Stage 6: Harvest & SUB transfer
The expanded seed is harvested in closed mode and aseptically transferred via sterile welding into the stirred production single-use bioreactor, completing the seed-train step.
